It has been reported that histological specimens shrink and/or expand during the formalin fixation process and preparation of histopathological specimens. In this study, to quantify the dimensional changes of histological specimens, we investigated the size of hard and soft tissues during the following processes: formalin fixation (studyⅠ), and from post-trimming to completion of the histopathological specimen (studyⅡ).<br>StudyⅠ consisted of soft tissue samples from 15 tongue resected patients, and hard tissue samples from 15 segmental mandibular resected patients. The dimensions were measured before and after formalin fixation. The mean shrinkage in soft tissues and hard tissues were −0.40% and 2.79%, respectively. The larger the surgical soft tissue specimen, the significantly smaller was its mean shrinkage.<br>StudyⅡ consisted of the soft tissue samples from 67 tongue resected patients, and hard tissue samples from 19 segmental mandibular resected patients. The dimensions were measured at the time of post-trimming and after completing the histopathological specimens. The mean shrinkage in soft tissues was 6.81%, compared to 9.35% in hard tissues. The mean shrinkage in hard tissues was significantly higher than that in soft tissues. In contrast to studyⅠ, studyⅡ showed that as the size of soft tissue specimen increased, its mean shrinkage also increased significantly.<br>These results provide useful feedback from histopathological diagnosis to clinical diagnosis, and help to choose more appropriate diagnosis and treatments.
