<p>Mechanical signals in the extracellular environment are transmitted to the cell nucleus through the actin cytoskeleton. The signal transduction mechanism via the actin cytoskeleton is considered to play an important role in regulation of cell homeostasis, but the details have not been clarified. In this study, we aimed to establish the method for the evaluation of the dynamics of actin cytoskeleton and nucleus based on live imaging of the three-dimensional morphology of the actin fibers and the nucleus in order to understand their mechanical interaction. The observation of the three-dimensional morphological changes of the actin fibers and the nucleus demonstrated that the pseudopod containing actin filaments locally extended and contracted, and the nuclear height and projected area changed with the passage of time. In addition, the 2D digital image correlation analysis suggested non-uniform deformation of the nucleoplasm. This intranuclear deformation is thought to be caused by the forces transmitted from the actin to the nucleus. This method can be utilized for correlation analysis to provide quantitative and mechanistic insights about the actin-nucleus interaction, during the osteogenic differentiation of mesenchymal stem cell.</p>
