Abstract
<p>This study was conducted with the aim of linking saliva tests to oral health guidance methods. Focusing on the oral cavity protective action of saliva, we measured the inhibitory activity on cysteine protease (papain), which is a proteolytic enzyme. We further examined the expression of cystatin S exhibiting this action and salivary protein concentration, and we investigated the relationship.</p><p>Whole saliva at the time of stimulation by ume candy drops was collected from 10 women, including 5 major students and 5 faculty members of N Junior College, and the inhibitory activity on papain (papain inhibition) and protein concentration were measured. Their salivary proteins were separated using sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and the expression level of cystatin S was measured based on the intensity of bands on western blotting.</p><p>As a result of measuring the papain inhibition and protein concentration of each whole-saliva sample, a positive correlation (correlation coefficient r = 0.724, p < 0.05) was observed. A positive correlation (r = 0.776, p < 0.01) was also observed between the results of western blotting with anti-cystatin S serum and papain inhibition. However, there was a correlation between the expression of cystatin S and protein concentration (r = 0.550), but this was not significant.</p><p>In this study, positive correlations were shown between papain inhibition and protein concentration, and between papain inhibition and the expression of cystatin S. By measuring these factors, it may be possible to estimate the individual oral protective effect and use it as an index for oral health guidance.</p>
