Abstract

<p>[Purpose] </p><p>To evaluate correlation performance of Bone marrow mode of Sysmex XN-V software, we use FCM for reference method and ratio of myelocyte and erythrocyte as evaluating parameters. </p><p>[Method] </p><p>We administrated 100 mg/kg of 5-Fluorouracil (5-FU) via tail vein to male 7 weeks age of Crl:CD(ICR) mice and Crl:CD(SD) rats and bone marrow specimen were obtained from femur on 0, 3, 5, 10 and the 14th day(mice) and on 0, 3, 7, 10 and the14th day(rats) after 5-FU administration as measurement samples. As cell marker, anti CD71 antibody (for Erythrocytes) and anti CD11b antibody(for myelocytes) were used for FCM measurement and calculated each erythrocyte and myelocyte ratio.</p><p>[Result, Consideration]</p><p>The result showed coefficient of correlation was more than 0.85 between Sysmex XN-V bone marrow mode and FCM method. This result was good correlation, however, several deviated data caused by following reason were also observed. In mouse data, deviation was observed in samples of 3 and 5 days after administration and those samples were considered highly affected 5-FU myelosuppression effect. These samples were very small cell number, therefore even tiny cell number difference caused big change in calculation of cell ratio. In rats data, deviation were observed in samples of 7 day after administration. These samples showed different dotplot position compared to other samples and this difference was considered cause of this deviation. </p>
