Rapid and sensitive determination of proteins in biological samples by FIA.

Abstract
A method for the determination of proteins in biological samples was developed by combining FIA with a Coomassie Brilliant Blue G-250(CBB G-250) dyebinding method. This system consists of two delivery pumps of reagent solution and carrier solution, a sample injector, a reaction coil and a spectrophotometer. The wavelength of the spectrophotometer was set at 600 nm and 0.1 m of reaction coil(1 mm i.d.) was used. Each flow rate of the reagent solution and carrier solution was 1.0 ml/min. The concentration of 60 μg/ml of CBB G-250 and 2.7% of perchloric acid in the reagent solution gave the maximum sensitivity. In the carrier solution 20 μl of the sample was injected. A linear calibration curve was obtained from 40 ng up to 1.0 μg of bovine serum albumin (BSA) as a standard substance. The minimum detectable amount of protein was 40 ng and the relative standard deviation for 1.0 μg of BSA (n=10) was 0.2%. This method allowed the analysis of 60 samples/h. Proteins in rat liver microsome were determined by this method. The analytical results showed good correlations with those by the manual method using CBB G-250 and by Lowry's manual method.
