A sequence-controlled polarograph, with a static mercury drop electrode (SMDE) was constructed using a programmable controller as a central processing unit (CPU). This polarograph permitted automatic operations in voltammetric measurement, e.g., the deaeration of test solution, the standing still, the formation of mercury drop electrode, the exporsure of the electrode to the test solution for a definite time interval at a constant or programmed potential, and the application of the potential sweep to the electrode. The sequence-controlled polarograph with SMDE has the advantages of automation and a minimum consumption of mercury. High reproducibility is predicted even when dynamic adsorption process is involved in the electrode reaction.As a test system for this work, trace analysis of protein using Brdicka current, the catalytic current produced by proteins (Brdicka-active proteins such as SH/SS-and heme-proteins) at mercury electrode in the presence of cobalt salt was investigated. The Brdicka current of bovine serum albumin (BSA) Was proportional to the concentration in the range between 20 and 200ng/ml with a relative standard deviation (R. S. D.) of 6.3% by the linear sweep voltammetry, and between 6 and 200ng/ml with R.S.D. of 4.9% by the differential pulse polarography. The linear calibration curves of BSA was not interfered with the presence of Brdicka-inactive protein such as gelatin in less than 2μg/ml.
