Recently, there has been an increasing number of reports on cancer therapies such as photodynamic therapy and hyperthermia. However, there is a problem that the therapies can only be used for superficial thin tumor cells. In this study, we synthesized novel porphyrin having thiol group which can be adsorbed onto gold surface in order to obtain porphyrin-modified gold nanoparticles. We think that heat is generated around tumor cells in a deep part of a human body by the irradiation of electromagnetic wave to the gold nanoparticles. Gold nanoparticles are more stable than other nanoparticles, because of superior stability against oxidation and less toxicity. Porphyrin takes a role as a protective agent of gold nanoparticles, and is expected to promote the selective attachment of gold nanoparticles to tumor cells. The nanosize of the gold nanoparticles was accomplished by the sodium borohydride reduction method and citrate reduction method. The combination of thiol and gold nanoparticles was confirmed by the quenching of porphyrin in the fluorescence spectrum and shift of the Soret peak in the UV-vis spectrum. We evaluated the characteristics of porphyrin-modified gold nanoparticles by the measurement of the residence time in blood, permittivity, and cytotoxicity test.
