An examination was made on the in vitro antimycotic activity of terbinafine (TBF), a new allylamine antimycotic, against 40 strains of Candida albicans freshly isolated from patients with superficial candidiasis. The study was carried out using the agar dilution method and the activity of TBF was compared with that of bifonazole (BFZ), an known imidazole antimycotic. The MIC values of TBF against these clinical isolates distributed over a wide range between≤0.16 and >80μg/ml with the geometric mean of 13.195μg/ml, while those of BFZ were from 1.25 to 10μg/ml with the geometric mean of 1.708μg/ml.<BR>The sub MIC values of TBF were determined by the micro-broth dilution method using a yeast nitrogen base medium (Difco) supplemented with 1% glucose, against 6 C. albicans isolates. The mean values of IC50 and IC90 of TBF were 0.46 and 3.25μg/ml respectively, whereas those of IC50 and IC90 of BFZ were 1.37 and 3.05μg/ml, respectively. These results suggest that TBF has a wider sub MIC zone against C. albicans than does BFZ.<BR>Subcultures of 8 clinical isolates were made on agar medium containing TBF to examine the possible development of TBF-resistance. The TBF-sensitivity of all tested isolates did not lower more than one tenth of the initial level after 20 subcultures.
