Light-microscopic, scanning electronmicroscopic, and transmission electron microscopic observations were made on the effect of intra-tympanic drug injection on the full-grown otoconia as well as on the pathogenesis of otolithiasis, by using rats and guinea-pigs. The results were as follows.<br>The findings in rat embryos of 16 gestational days revealed aggregated substances of a high electron density, which were of stratified structure, namely, a precursor of otolitn just on the sensory epithelium of the incomplete macula. Furthermore, the substances increased in number and volume, traveled gradually, upward and formed the so-called, “cupular zone” till they became full-grown otoliths. (or a full-grown otolith)<br>When aminoglycoside, chloramphenicol succinate and 7W/V% Sodium Bicarbonate were intra-tym-panically administered to guinea-pigs, changes were seen not only in the sensory cells but also in otoliths, together with their displacement and decrease in size. Especially, the injection of chloramphenicol succinate resulted in the phagocytosis of otoliths by the supporting cells.
