The effects of different media and some phyto-hormones on both the growth and the rooting in shoot tips culture of cv. Yabukita were investigated.<BR>1) In the eight kinds of media with different compositions of macronutrient without phyto-hormones, the growth of tea shoot tips in the NITSCH'S, GAMBORG'S or MURASHIGE & SKOOG'S medium was better than that in the other media.<BR>2) The growth of tea shoot tips was obviously different among the several kinds of cytokinins in the MURASHIGE & SKOOG'S medium. In the case of kinetin, benzyladenine (BA), adenine, zeatin and isopentanyladenine, the growth of tea shoot tips was accelerated slightly by the addition of 3.0mg/l of each cytokinins, as compared with 0.1mg/l of each one.<BR>3) When BA was combined with various concen-trations of naphthalene acetic acid (NAA), callus formation on the base of shoot tips was accelerated, however the growth was inhibited in the NAA added plots. The combination of BA and gibberellin (GA) accelerated the growth of shoot tips. Particularly, the combination of BA (0.1 or 1.0mg/l) and GA 5.0mg/l was better than the other combinations in growth of shoot tips.<BR>4) The ideal conditions of root induction from the shoot in vitro were the liquid medium, containing a half or a quarter strength of macronutrient of MURASHIGE & SKOOG'S medium which was supplemented with 0.5-1.0mg/l of indole butyric acid (IBA).
