Human burst-forming unit-granulocyte (BFU-G) derived colonies which composed of neutrophils were formed in culture with stem cell factor (SCF), interleukin-3 (IL-3), granulocyte colony-stimulating factor and erythropoietin. The BFU-G colonies were very large in size and composed of multiple subcolonies, which resembled burst-forming unit-erythroid (BFU-E) derived colonies in morphology; but their hue was non-red, white or grey. The BFU-G colonies may be derived from more primitive granulocytic progenitors, which have high proliferative potentials, than colony forming unit-granulocyte. This study suggests that SCF and IL-3 stimulate proliferation of primitive granulocytic progenitor cells, and promote growth of BFU-G colonies.
