A simple micro-analysis of benzo(a)pyrene (BaP) in air-borne particulates was studied. Air-borne particulates were collected on glass-fiber filters (20×25, cm) by hi-volume sampler. A circle of 15 mm ir diameter was cut out from each glass-fiber filter by a belt-ponch, and the circle was used as a sample in BaP analysis in this method. These samples were attached to kieselguhr layer of dual band thin-layer chromatoplate, which composed of kieselguhr layer (5×20, cm) and 26% acetylated cellulose layer (15×20, cm). Polynuclear aromatic hydrocarbons like BaP in the sample was extracted on the kieselguhr layer by continuous development with ethyl ether for 20 minutes in a dark place. The plate was then run with methanol-ethyl ether-water (4:4:1, v/v) in order to separate BaP on the 26% acetylated cellulose layer. The development was continued until the solvent traveled 10 cm on the 26% acetylated cellulose layer. The BaP spot separated on the thin-layer was scraped off into a small centrifugal-tube. After adding 5 ml of dimethyl sulfoxide (DMSO) into the tube, ultra sonic extraction was carried out for 10 minutes in order to dissolve BaP completely. The extracted DMSO solution was centrifuged for 5 minutes. BaP in supernatant was identified by comparison of its fluorescence and exitation spectra with those of standard BaP solution and was determined spectrofluorometrically by means of the narrow base line method. Recovery of BaP and its coefficient of variation in this method were 99.9% and 3.3%, respectively. This method was highly sensitive so as to be able to determine accurately BaP at the level of 1 ng or less. Furthermore, BaP data for the same particulate samples analyzed by the both method, the present method and the method which consisted of ultra-sonic extraction, one-dimensional dual band thin-layer chromatography, and spectrofluorometrical determination were in good agreement. It was also proved that distribution of BaP in the glass-fiber filter was nearly uniform. Therefore, it is concluded that this method is useful to determining BaP in air-borne particulates.
