Description

The respective determination of semi-micro amounts of D-glucosamine hydrochloride and its four derivatives; N-methyl-D-glucosamine hydrochloride, N-acetyl-D-glucosamine, tetra-O-acetyl-D-glucosamine hydrochloride, pentaacetyl-D-glucosamine has been investigated.
The recommended procedure is based on the reduction of alkaline cupric sulfate and the determination of the separated cuprous oxide, dissolved in nitric acid, by complexometric titration of resulted cupric ion with ethylenediamine tetraacetic acid disodium salt with murexide as an indicator.
The calibration curves are made between the amount of cupric ion and that of these five compounds (Fig. 3 and Fig. 4). In the case of D-glucosamine, the curve is linear over the range of 110 mg of sample.
The effect of coexisting metal ions on the determination of D-glucosamine hydrochloride is shown in Table II and Table III.
As for the reduction powers of these five compounds against Fehling reagent, the following order has been obtained: N-methyl-D-glucosamine≥D-glucosamine>tetra-O-acetyl-D-glucosamine>N-acetyl-D-glucosamine≥pentaacetyl-D-glucosamine.
