Following conditions were studied in order to improve a decolorization of phenolphthalein in alkaline media: species, concentration and pH of buffer solutions, standing temperature and time, foreign compounds, etc. As a result, the following standard procedure was chosen for the determination of phenolphthalein: fifty mg of phenolphthalein is dissolved in 100 ml of ethanol and the solution diluted with ethanol to yield (50500) μg/5 ml of the sample. Five ml of the sample solution is taken being evaporated to dryness on water bath. The residue is dissolved in 0.1 M carbonate buffer(pH 10.6) to give exactly 50 ml, and then the absorbance at 555 nm is immediately measured at 20°C (reference: the buffer solution). 0.1 M carbonate buffer(pH 10.6) gave better result than 0.1 M glycine buffer(pH 11.3) and 0.1 M phosphate buffer (pH 11.3) which have been widely used.
