The flow injection/chemiluminescence system with detectability at a concentration of 2 × 10-6 M for 20 μl injection of hydrogen ion was successfully applied to quality control of roasted coffee beans. The chemiluminescent reaction was composed of didodecyldimethylammonium bromide bilayer vesicle/ Eosin Y/ hydrogen ion/ sodium hydroxide. The relationship between the chemiluminescence intensity (X=log A where A is amperes) and the concentration (Y=log M where M is molarity) for hydrogen ion in the coffee essence was linear in the range of concentration from 6.8 × 10-6 M to 2.5 × 10-3M (Y= 0.73X+2.29). The determination was also selective and the reproducibility in terms of relative standard deviation was less than 1.1% for a 8.3 × 10-6 M hydrogen ion injection (10 repeated tests). The chemiluminescent signal originating in the extracted essence from roasted coffee beans was readily applicable to roast-process control and quality control of the coffee beans. Control checks were very easily and rapidly performed by the proposed flow system. The separation and identification of key-materials relating to the chemiluminescence signal are in progress.
