Avenanthramides, a series of substituted N-cinnamoylanthranilate derivatives, are phytoalexins in oats. Hydroxycinnamate and anthranilate are derived from the shikimate pathway which branches at chorismate. Chorismate mutase (CM) is the first enzyme of the branch that leads to phenylalanine and phenylpropanoid biosynthesis. The induction of CM was investigated in primary oat leaves using oligo-N-acetylchitooligosaccharides as elicitors. The CM activity started to increase 6 hr after elictitation and reached a maximum by 9 hr. Among the oligo-N-acetylchitooligosaccharides tested, tetra-, penta-, and hexasaccharides effectively induced the enzyme activity in a dose-dependent manner. The activity was separated into two major peaks by anion exchange chromatography indicating that CM consists of at least two isoforms. Comparison of untreated and elicitor-treated leaves revealed that only one CM isoform was responsive to the elicitor. These results suggest a regulatory role of CM in avenanthramide biosynthesis.
