72. Isolation and identification of novel β-Yariv reagent-reactive arabinogalactan-proteins from aleurone layers of rice (Oryza sativa)

Arabinogalactan-proteins (AGPs) are extracellular glycoproteins playing important roles in plant growth and development. We have previously reported the possibility that AGPs are involved in the induction of α -amylase by gibberellin in barley aleurone layers by using the Yariv reagent (β -glucosyl Yariv) that has been presumed to specifically bind with AGPs. We have isolated several Yariv reagent-reactive proteins from rice bran rich in aleurone cells. The N-terminal sequences of OsAGP1 and OsAGPEP1〜3 have been determined from hydrophilic fractions obtained by RP-HPLC. A novel non-specific lipid transfer protein-like protein (OsLTPL1) and a novel early nodulin-like protein (OsENODL1) were also identified in the more hydrophobic fractions from RP-HPLC. A Yariv reagent-reactive tryptic fragment obtained from the OsLTPL1 fraction was identified to be a fragment of OsLTPL1 having typical AG glycomodules, which supports that the OsLTPL1 is really reactive with Yariv reagent. Since no Yariv reagent-reactive fragment was identified in the digest of the OsENODL1 fraction, this fraction was further purified with successive Yariv reagent precipitation and additional gel permeation-HPLC. The purity of the OsENODL1 fraction provides conclusive evidence that OsENODL1 was reactive with Yariv reagent. An expression analysis of the genes coding for these proteins was also performed. While OsAGP1, OsAGPEP1〜3 and OsLTPL1 were expressed in various parts of rice, OsENODL1 showed temporally and spatially specific expression in the aleurone layers.
