Abstract
The K2 type of killer plasmids (dsRNAs) was transferred from a karyogamy-defective haploid strain 5170 into an industrial diploid alcohol yeast M-111 through protoplast fusion to breed a killer alcohol yeast K2-M-111. The DNA content of strain K2-M-111 thus bred was at the tetraploid level.<br> The fermentation ability of strain K2-M-111 was several-fold greater than that of strain 5170, and the same as that of strain M-111. Strain K2-M-111 produced the same level of byproducts from molasses as that produced by strain M-111.<br> Strain K2-M-111 effectively killed MCA-111 cells inoculated as contaminants during fermentation of molasses at 30°C and pH 3.2_??_4.2. The killer activity observed in the continuous fermentation with immobilized cells at pH 5.2 was significantly higher than that in batch-wise fermentation. The killer activity, alcohol production and attenuation of sugars of strain K2-M-111 were confirmed to be stably maintained for 65 days even when the retention time was changed from zτ=10 hr to τ=6 hr in continuous fermentation.
