Mammalian cell culturing has become a new technology of bioindustrial scale for preparation of such cellular products as monoclonal antibodies. Serum-free culturing of cells is an important step in the process. A new basal serum-free medium is described here for the culture and growth of immunoglobulin-secreting hybridomas. In some cases the new basal medium (e-RDF) supplemented with insulin, transferrin, ethanolamine and selenite (ITES) produced two to three times higher murine myeloma and lymphoma cell densities than conventional basal medium (c-RDF) supplemented with growth factors of 10% fetal calf serum. However immunoglobulin productivity per cell of some hybridoma lines was similar irrespective of the type of media. Depositions of immunoglobulin in culture medium were in proportion to the cell density in the medium. Therefore, growing hybridomas in higher density is one way to obtain a larger amount of monoclonal immunoglobulin molecules in vitro. More immunoglobulin was deposited in e-RDF supplemented with ITES than in c-RDF supplemented with ITES or serum by factors of 2.8 or 1.6 for 5F9-10C cells and 2.0 or 1.5 in case of 4C10B6 cells. Greater immunoglobulin production by hybridoma cells appears possible by improving the cell culture system using the e-RDF medium.
