Photoyellowing and photodegradation of silk fibroin are commonly accepted to be a radical process. Free radicals play an important role in the mechanism of photoyellowing and photodegradation. When silk fibroin is irradiated in vacuo at 77°K (-196°C) with ultraviolet-light, a double spectrum is observed. The spectrum is attributed to a glycyl radical formed by hydrogen abstraction from Cα position (splitting constant 19 G. g-value 2.0031). Glycyl radicals of U. V.-irradiated silk fidroin in vacuo react with oxygen promptly, when exposed to air at room temperature, and were converted to peroxy radicals. It means that oxygen molecules diffuse easily in silk fibroin. A singlet and doublet spectra are observed immediately after U. V.-irradiation of silk fibroin is placed in air. A concentration of peroxy radical decreases with the increases of humidity in atmosphere. It seems to mean that peroxy radicals decay quickly in high humidity, because the molecular motion of silk fibroin is accelerated by water. A mechanism of the photoyellowing and photodegration of silk fibroin is discussed.
