This study was undertaken to evaluate the methods of wing disc culture in the fifth larval instar of Bombyx mori, and to observe the mode of differentiation of the wing disc in culture. The best results were obtained when the strainless mesh method was employed among the three types of organ setting methods tested. The higher the O2 partial pressure, the faster the wing disc growth progressed. Complete differentiation including scale formation was even observed under ambient air conditions. The addition of 10% Bombyx serum was necessary for the complete differentiation, which was also observed in the medium containing 0.02-0.1μg/ml ecdysterone. The mode of differentiation of the wing discs of the larvae on days 1, 6 and the prepupa was identical, and mitoses, cuticle deposition and scale formation were observed in each case.
