Description
1. All isolates of Diaportahe nomurai (D1, D3) and Thyrostroma mori (T1, T2) need thiamine as a nutrient. The optimum concentration of thiamine for mycelial growth of those fungi was about 5μg/l.
2. The requirement to carbon source of D1 was similar to that of T1. The two pentoses, L-arabinose and D-xylose, supported a good growth of those fungi. Two isolates showed a good growth on the four hexoses, D-glucose, D-mannose, D-galactose, and D-fructose. Maltose and saccharose also gave a good growth for two fungi. However they made a poor or no growth with L-rhamnose and lactose. Considering polysaccharides, a relatively large amount of mycelium with those fungi was produced by citrus pectin, dextrin, and soluble starch. But they were unable to utilize inulin and carboxymethyl-cellulose. Glycerol, D-mannite, and D-sorbite were poorly utilized by those fungi. A little growth of D1 was brough by an administration of succinic acid. This isolate did not show any growth on citric acid, lactic acid, acetic acid, and oxalic acid. The mycelial growth of T1 was inhibited in media containing any one of five organic acids as the sole source of carbon.
3. Isolate D1 grew well on most of amino acids tested, showing maximum growth with L-aspartic acid, L-asparagine, and glycine. Isolate T1 grew well on L-asparagine, L-proline, L-glutamic acid, and L-aspartic acid. However, T1 showed a little growth on L-arginine HCl, and a poor or no growth on the rest of amino acids tested. Isolated D1 showed a good growth on potassium nitrate, sodium nitrate, and ammonium phosphate (dibasic), a little growth on ammonium sulfate, and a poor or no growth on ammonium nitrate and sodium nitrite. On the other hand, isolate T1 showed a poor or no growth on each of six inorganic nitrogen sources tested.
