Abstract

Both sericins extracted from the silkgland and the cocoon layer from Nds silkworm were studied by means of X-ray, DTA, infrared spectroscopy, and ultramicroelectrophoresis. The following results were obtained:<br>1) The isoelectric points of both sericins extracted from silkgland and cocoon layer from Nds silkworm were pH 4.05, and they did not differ remarkably with that of sericin prepared by MOSHER method.<br>2) The solubility of sericin in cocoon layer from Nds silkworm was similar to that of normal cocoon layer.<br>3) IR-spectrum of sericin extracted from the silkgland in Nds silkworm was much the same as that of sericin prepared by MOSHER method.<br>4) X-ray diffraction pattern of sericin extracted from the silkgland in Nds silkworm was coincided with that of sericin prepared by MOSHER method except interference of 8.85 A (side chain spacing). Also, X-ray diffraction pattern of bave from Nds silkworm was circular showing some fibre diagram. That is, the interference of 4.57A (back-bone spacing) was appeard strongly about the equator, and it means that considerable polypeptide chains of sericin were oriented pallarel to fibre axis.<br>5) Thermogram of sericin extracted from the silkgland in Nds silkworm had double endothermic peaks at 232°C and 272°C. The decomposition temperature was higer than that of sericin A prepared by MOSHER method, but similar to sericin B.
