Abstract

Differences between properties of sericins prepared from cocoon layers of B. mori reared with mulberry leaf and artificial diet, were studied by means of ultramicroelectrophoresis, X-ray diffraction, DTA and IR-spectroscopy.
The following results were obtained:
1) In regard to this experiment, the content ratio of sericin in cocoon layer of B. mori reared with artificial diet was a little less than that of mulberry leaf. Also, sericin A which is hydrophilic protein, was contained much more in cocoon layer of B. mori reared with mulberry leaf than that of artificial diet, simultaneously the former had higher solubility than the latter.
2) In isoelectric point (pH 3.7-3.8), IR-spectrum and X-ray diffraction patterns, no significant difference can be found between sericins of cocoon layers of B. mori reared with artificial diet and those of mulberry leaf.
3) Thermograms of sericins extracted from cocoon layers of B. mori reared with artificial diet and mulberry leaf had similar patterns, but the decomposition temperatures of the former were higher about 3-9°C than those of the latter. In particularly, its difference of sericin A of inner layer was remarkably large.
4) In freeze dried sericin from the precipitate, it was observed that the absorption band of amide I had two peaks about 1630cm-1 and 1660cm-1, and that endothermic peak at decomposition was increased remarkably.
