Abstract

A mannanase was purified from extracts of bran Koji culture of Aspergillus niger van Tieghem sp. by column chromatography of DEAE-Sephadex and Sephadex G-75, electric focusing method, and zone electrophoresis.<br> The pH optimum of purified mannanase was 3.5, the pH stable range was 4_??_7, and the thermostable range was below 50°C.<br> When the mannanase reacted to galactomannan from soy bean hull, galactomannan from guar gum, mannan from coffee bean and glucomannan from arum root, mannooligosaccharides were found and the hydrolytic limit to those substrates were 12, 12, 58, 39% respectively.<br> So this enzyme was considered as to be endo-β-mannanase.
