It is often difficult to identify whether cells in body cavities of cancer patients are cancer cells, mesothelial cells, or macrophages-We made successive observations of such cells with light microscopy, scanning electron microscopy, and transmission electron microscopy.The body cavity fluids of cancer patients were centrifuged with an autosmear and fixed in 1% glutaraldehyde. After Papanicolaou staiing, the target cells were photographed.At this time, a few drops of quetol651, an aqueous resin, were added to the stained samples on the laboratory-made film slide.The film slide was then immersed in 90, 80, 70, 50, and 30 % quetol aqueous solutions, and distilled water for10minutes each, to remove the quetol.Thereafter, it was routinely double-fixed for electron microscopic examination.The specimen on the film slide was observed and photographed with a scanning electron microscope.This evaporated specimen was immersed in a graded series of ethanols for10minutes each, and hydrated in phosphate buffered saline for 30 minutes.After dehydration and epon-embedding, ultrathin sections were observed with a transmission electron microscope and photographed.There was little difference in the observations of the cytoplasm between cells fixed in 95% alcohol for ordinary Papanicolaou staining, and 1% glutaraldehyde fixed cells.The specimen could, however, be satisfactorily evaluated for cytodiagnosis.Scanning electron microscopic observation clearly showed the fine structure of the cell surface, and transmission electron microscopic observation definitely showed the preserved inner structure of the cell In addition to the Papanicolaou staining, the electron microscopic findings were very useful for distinguishing among the above three types of cells.
