Five steps of motility were classified in protozoa observed in five fields of the microscope. They were converted into indices. This method was applied to 40 healthy dairy cows in the field. As a result, the activity index was 4.8 for the genus Isotrich, 4.7 for Entodinium, and 4.7 on the average for Diplodinum, Ophryoscolex, and Epidinum. The degree of activity of each protozoon decreased with the lapse of time after collection. The degree of decrease varied with the genus.<BR>When the rumen juice was filtered by the double gauze layer method, a considerably large number of protozoa, together with the residue of the juice, were attached to the gauze. This phenomenon had influence on the distribution of protozoa.<BR>For dilution and staining of the rumen juice, MHS solution was superior to MHB solution in staining property. Both solutions were used for counting to prepare two-to tenfold dilutions of the rumen juice. There was no significant difference in the count of protozoa between the use of Fuchs-Rosenthal's chamber and that of a plankton chamber.
