Abstract
Black bass were unofficially introduced into the Tsuga dam reservoir in the Shimanto River basin and the Sameura dam reservoir in the Yoshino River basin, Kochi prefecture, Japan. We identified the species and estimated the genetic variability of these two populations using mitochondrial DNA and microsatellite DNA analysis. Based on mitochondrial DNA control region sequence analysis, both populations shared the same haplotype (c) of largemouth bass, Micropterus salmoides salmoides. Based on microsatellite DNA analysis of 7 loci, the genetic variability of each population was similar in terms of both the number of alleles per locus and heterozygosity. Moreover, we observed significant genetic differentiation between the two populations (pairwise FST; P<0.01), suggesting these populations may have a different origin. Our results provide a baseline against which to monitor the range expansion of black bass in Kochi prefecture.
