Rainbow trout (mean body weight 21.1 g) and crucian carp (mean body weight 3.92 g) were fed test diets for 114 and 61 days, respectively. In trout, the fillet fat, 20:5n-3, and 22:6n-3 contents were, respectively, 1.5, 1.7, and 1.4 times higher (P<0.05) in fish fed a low-phosphorus diet (LP, 0.23% P) than those fed a high-phosphorus diet (HP, 1.06% P). In carp, the fillet fat, 20:5n-3, and 22:6n-3 contents were, respectively, 1.7, 1.6, and 1.3 times higher (P<0.05) in fish fed the LP diet than those fed the HP diet. However, when fish oil in the LP diet was replaced with lard, the fillet fat content did not increase. To investigate the molecular mechanism of body fat accumulation under dietary P restriction, the expression of 13 genes in the trout liver was determined by RT-QPCR. The mRNA expression of CPT1 (carnitine palmitoyltransferase-I), a rate-limiting enzyme in lipolysis, tended to be lower in the LP than HP groups. The expression of ACC1 (acetyl-CoA carboxylase), a rate-limiting enzyme in lipogenesis, tended to be higher in the LP than HP groups. The altered metabolism by chronic P restriction appears to be lipogenic, ketogenic, and possibly thermogenic.
