- Evaluation of Environmental Estrogenic Activities in the Yangtze River Estuary: Utilization of Vitellogenin Production in Red Lip Mullet

The objective of the present study was to survey environmental estrogenic activities in three different estuary areas around the mouth of the Yangtze River, China. In this survey, we utilized production of vitellogenin (Vg), a major estrogen-sensitive, liver-derived serum precursor of egg yolk in red lip mullet (Chelon haematocheilus) as a marker for the detection of estrogenic activities; both circulating and hepatic expression levels of a major Vg subtype (VgB) were quantified by a chemiluminescent immunoassay (CLIA) and a real-time quantitative reverse transcription PCR (qPCR), respectively. Results revealed that VgB (both protein and mRNA levels) was non-detectable or barely detectable in fish caught from surveyed areas; these levels were quite similar to levels obtained for fish acclimated to an artificial estrogen-free environment (control fish). In addition, any gonadal abnormality was hardly confirmed in these fish by histological observation, although one exceptional case (one out of 40 juvenile individuals) indicated formation of a testis-ovum. These results suggested that surveyed areas were not at serious risk of estrogenic activities.
