Abstract

Circulating Gal, GalNAc-glycoproteins were determined in various liver diseases and carcinoma of digestive organ by using inhibition test of hemagglutination with RCA-1 and RCA-2 lectins which are specific for Gal and Gal, GalNAc respectively. Both RCA-1 and RCA-2 hemagglutination inhibition titers were elevated in hepatocellular carcinoma, liver cirrhosis and carcinoma of digestive organ, while they did not correlate to serum AFP nor CEA values. In chronic liver disease, they partially correlated to cholinesterase and KICG but not in hepatocellular carcinoma nor carcinoma of digestive organ. Therefore the increase of Gal, GalNAc-glycoproteins seems to originate from carcinoma tissue as well as liver injury which increases circulating asialoglycoproteins.<BR>160ml (2.7g protein) of ascites from a patient with hepatocellular carcinoma were applied to RCA-2 agarose affinity chromatography and antiwhole human serum-Sepharose 4B affinity chromatography by which serum components were absorbed. After then 26.2mg p otein of Gal, GalNAc-glycoproteins were obtained. SDS-polyacrylamide gel (7.5%) electrophoresis of these glycoproteins showed 3 main bands (64K, 46K, 15K) and 2 sub-bands (250K, 210K).
