The changes in the intrahepatic portal vascular resistance during prolonged infusion of epinephrine were studied in the isolated perfused normal and cirrhotic rat livers. Liver cirrhosis was produced by injections of carbon tetrachloride, and the mean portal vein pressure was 196mm H2O (normal: 112mm H2O). The intrahepatic portal vascular resistance in the cirrhotic liver increased rapidly and reached a peak within 5 minutes after infusion of epinephrine. The peak of the intrahepatic portal vascular resistance in the cirrhotic liver was larger than that in the normal liver at the same concentration of epinephrine. Furthermore, the maximum increase in the intrahepatic portal vascular resistance in the cirrhotic liver obtained at lower concentration of epinephrine as compared with the normal liver. These findings suggest that severer portal hypertension is resulted in the presence of vasoconstrictive substances in the portal blood in liver cirrhosis, and that a rapid increase in vasoconstrictive substances in the portal blood may lead to the rupture of esophageal varices.
