Our previous reports have made clear that glucagon and insulin stimulate the liver regeneration by their synergistic action in experimental and clinical trials.<BR>In this paper we aimed to reveal interaction of their pancreatic hormones to molecular levels in the damaged liver.<BR>Rats were administered of d-galactosamine (1000mg/kg body weight) by intraperitoneal injection. After 24 hours we treated with administration of glucagon (1mg/kg body weight) and regular insulin (1unit/kg body weight).<BR>These rats were divided into three groups as the followes; glucagon-insulin group, glucagon group and control group.<BR>After studying of the biological parameters for the liver regeneration, we revealed positively the localization of alpha-fetoprotein, cyclic AMP by immunofluorescence method in liver tissue and DNA synthesis in vivo by autoradiography of 3H-thymidine.<BR>Survivors were significantly enhanced with administration of glucagon and insulin in comparison to glucagon alone and control.<BR>It is concluded that administration of glucagon and insulin simultaneously stimulate the biological parameters and improve the survival rate.
