Effects of Cyclic Total Parenteral Nutrition on Circadian Rhythmicity of Serum Parameters, Organ Weight and Histological Findings of Liver, Kidney, and Lung in Rats.

Circadian rhythms of serum parameters, organ weight and histological findings of liver, kidney, and lung were investigated in rats receiving cyclic total parenteral nutrition (TPN) and continuous TPN. Results showed that rhythm patterns for body weight and liver weight were similar between cyclic TPN and control groups (rat chow, ad libitum), whereas no circadian rhythms of body weight and liver weight were observed in the continuous TPN group. Circadian changes of kidney and lung weight were not significant and histological findings revealed normal architecture of kidney and lung in the cyclic TPN group, suggesting that cyclic TPN may not have an adverse effect on renal and pulmonary function. Serum parameters regarding the fat metabolism showed an obvious circadian rhythm in the cyclic TPN group, in which very low density lipoprotein (VLDL) was synthesized predominantly in the fed state and fatty acids from the peripheral tissues were re fluxed to the liver in the fasted state. On the other hand, in the continuous TPN group. VLDL synthesis was accelerated and reflux of free fatty acids was suppressed all the time. Histologic findings on the liver showed that hepatic steatosis developed in the continuous TPN group, whereas circadian fluctuation of glycogen deposit was recognized in the cyclic TPN and control groups. These results suggested that cyclic TPN induces a physiological circadian rhythm of carbohydrate and fat metabolism which may prevent hepatic steatosis.
