Description
Cefditoren pivoxil, a cephem antibiotic, has been widely accepted clinically. In this study, drug distribution to the brain was evaluated using the microdialysis method and the effect of cefditoren on brain tissue was investigated. The implanted microdialysis probes in the rat striatum and the jugular vein were perfused with Ringer's solution. A dialysate was collected every 20 min after administration of cef ditoren (10, 30, 100mg/kg i.p.) or of loxacin (New quinolon antibiotics, 30mg/ kg i.p.) . Cef ditoren and of loxacin concentrations in the dialysates were determined by the HPLC/ECD system. Cefditoren (10, 30mg/kg) levels promptly increased in both the brain and blood, reached maximum concentrations at 60 and 40 min, respectively and then decreased gradually until 5hr after administration. In the cef ditoren 100 mg/kg treated group, the blood concentration increased promptly and drastically even though the cef ditoren level in the brain increased slightly. Similar changes were observed in the of loxiacin treated group. In conclusion, the effect of cefditoren on the brain tissue is marginal since the extraceller concentration is quite low in an ordinary dose determined by the microdialysis technique.
