Guinea pigs were immunized with various sorts of renal tissue antigens obtained from rat, including the kidney suspension, glomerular basement membrane (GBM), cortical renal basement membrane and medullary tubular basement membrane (TBM), which were emulsified in complete Freund's adjuvant. <BR>A few weeks after the immunization, guinea pigs developed glucosuria. By lightmicroscopy, severe tubulointerstitial nephritis (TIN) was found in guinea pigs which were immunized with rat kidney suspension and cortical basement membrane. <BR>The pathological findings of the TIN were characterized by the tubular atrophy and the infiltration of mononuclear cells in the interstitial tissue. On the contrary, the glomeruli showedd almost normal or mild hypercellularity. <BR>By immunofluorescence, IgG and C3 of guinea pig were found in linear fashion along the TBM and IgG along the GBM prior to the development of light microscopical abnormality. <BR>Seven out of 10 sera or eluated obtained from the renal tissue of guinea pigs immunized with cortical renal basement membrane bound with both of TBM and GBM, but the other sera with TBM only. <BR>The experiment of passive transfer by means of sera containiug anti-TBM and anti-GBM antibody revealed that guinea pig IgG was stained in linear fashion along cortical TBM and GBM in the recipients, although no light microscopical abnormality was found in renal tissue. <BR>These data suggested that TBM was capable to have TBM specific antigenecity and also anti-TBM antibody may play a major role to the development of TIN.
