It is well known through the many reports published in the literature in Japan and abroad that there is an association between IDDM and HLA. As for the genetic mechanism of this association, the most predominant is the linkage disequilibrium theory in which HLA gene and gene regulating the development of IDDM are assumed to be linked with a marked linkage disequilibrium with specific HLA gene.<BR>However, studies have not been made on the estimation of coefficient of linkage disequilibrium. Selection, consanguinity, and migration are considered to be factors which influence linkage disequilibrium.<BR>The authors developed a method of estimating linkage disequilibrium which takes into consideration selection and consanguinity and analyses were attempted on the association of IDDM to HLABw 54 and DYT.<BR>(1) In these analyses the coefficient of linkage disequilibrium did not reach a significant level even when selection and consanguinity were not taken into consideration.<BR>(2) The effect of selection and consanguinity on linkage disequilibrium is large. It was also demonstrated that the possibility of deriving erroneous results would be large if this were not properly taken into account.<BR>Furthermore, in estimating linkage disequilibrium, considerations were made on the method of data treatment, on the question of estimation method, and on the effects of migration.
