Sulfonylurea drugs are widely used in the treatment of non-insulin-dependent diabetes mellitus. Recently, many investigators have come to believe that the extrapancreatic effect of sulfonylureas is more attractive in terms of the amelioration of insulin resistance in NIDDM. Despite many studies on the extrapancreatic effects of sulfonylureas, their mechanisms are poorly understood. Therefore, in order to provide some insight into such agents, we studied the effects of a new potent sulfonylurea drug, HOE490, on hepatic metabolism in the isolated hepatocytes of normal rats. The following results were obtained. 1) HOE490 caused significant suppression of hepatic gluconeogenesis from alanine, lactate, pyruvate, gly cerol and fructose. 2) In the presence of alanine and palmitate, HOE490 decreased hepatic ketone-body production significantly in a dose-dependent manner. 3) 500μEM HOE490 exhibited antigluconeogenetic effect similar to that of 1.5mM tolbutamide and 1 mM buformin. 4) The oxidation of [U-14C] alanine, [1-14C] glutamate, [1-14C] pyruvate, [1-14C] glucose and [1-14C] palmitate was significantly increased by HOE490-1, while tolbutamide did not display any such effects.<BR>Thus it appears that the inhibitory effect of HOE490 on gluconeogenesis and ketogenesis involves increased TCA cycle activ ity in a mechanism which differs from tolbutamide.
