Description

A method of separating human peripheral blood monocytes (HPBM) from white blood cell buffy coat by counterflow centrifugal elutriation (CCE) is described. An average of 8.7×107 HPBM, with a purify of >90%, were recovered from one unit of buffy coat processed from 450ml of whole blood. HPMB recovered by this method were viable as determined by trypan blue exclusion, had intact morphology according to light and electron microscopic observation and were not spontaneously tumoricidal as determined by a 3-day cytotoxicity assay. These HPBM could be activated to the tumoricidal state by either bacterial lipopolysaccharide (LPS) or recombinant interferon gamma (rIFN-γ)+muramyl dipeptide (MDP) and were phagocytic as judged by carbon particle or liposomal uptake. Their tumor cytotoxic function was comparable to that of HPBM separated by other methods.
