Abstract

In Japan, house dust plays a very important causative role in bronchial asthma. For this reason, six sera from house-dust-sensitive individuals were fractionated by DEAE cellulose column chromatography, and the localization of skin-sensitizing antibody was examined by the passive transfer test of Prausnitz and Kustner. It was demonstrated that thus isolated γG contained no detectable γA by the use of double gel diffusion with specific gout antisera, and the isolated γG showed no detectable skin sensitizing activity. A fraction which showed the highest skin-sensitizing activity contained γA, transferrin and tailing γG. The amout of γA in the fraction estimated by an immunoassay was smaller than other fraction which showed lower skin-sensitizing activity. A fraction which contained γM showed no skin sensitizing activity. On Sephadex G-200 the skin-sensitizing activity appeared with γA in eluates of intermediate portion between macro globulin peak and γG peak, in three cases. Other than skin-sensitizing antibody to house dust, skin-sensitizing antibodies to egg, candida or ragweed pollen in the sera of atopic patients, and further induced skin-sensitizing antibodies to ACTH and amidopyrine were examined by DEAE cellulose column chromatography. They showed similar situation in relation to immunoglobulin fractions as in the case of house dust allergy. These results suggest that skin sensitizing antibody, whenever it spontaneously occurred or it was artificially induced, is associated to γA more than to γG.
