The fluoride uptake from APF solution (pH 3.6, 0.9% F) applied to the enamel surface of deciduous teeth was measured. Enamel blocks of 30 specimens from deciduous teeth without carious lesions were used in this study. Five specimens were controls and APF was applied to the other specimens for 15sec, 30 sec, 60 sec, 120 sec, and 180 sec. Immediately after each application, the specimen was rinsed for 60 sec with distilled water. Six successive layers of enamel were etched away by exposure to 1.0 ml of 0.5 M perchloric acid. Fluoride was determined by a fluoride ion electrode and calcium by atomic absorption spectorophotometry.<BR>The results were as follows:<BR>1. The uptake of fluoride increased with increased application time from 15 sec to 180 sec, especially in the outermost layers.<BR>2. APF caused a statistically significant (P<0.01) increase of the fluoride concentration on the deciduous enamel surface during 15 sec (>5000 ppm F, 1.6μm) and 30 sec (>8000 ppm F, 0.8 μm).<BR>3. Fluoride gradients of the deciduous teeth were also determined using the formula Y=bxm [Y=F (ppm), X=depth (μm) ]. Using this formula, comparisons of the fluoride concentrations at the same depth between the samples were possible.<BR>4. The fluoride concentration (1600 ppm, 1775 ppm) found at 5 μm was about twice that of the control (870 ppm) in the groups in which the solution was applied for 15 and 30 sec, respectively. Furthermore, with applications of 60 sec or 180 sec, the fluoride was twice as high as in the controls up to a depth of 10 μm (about 1200 ppm). However, the fluoride concentration in the innermost layers did not differ from the control.
