The degradation of soluble and insoluble polysaccharides of human dental plaque and S mutans 6715 by Chaetomium gracile dextranase and the increase in the acid resistant property of bovine enamel caused by sodium monofluorophosphate (MFP) were examined to confirm their characteristic cariostatic properties. Moreover, effects of dextranase, MFP, and their combinations on the reduction of dental caries and plaque in rats were examined to estimate their additive cariostatic properties.<BR>Polysaccharides of human dental plaque and S mutans 6715 were degraded by 550 units/ml of dextranase. In particular, soluble polysaccharides were degraded about 75% and 55%, respectively.<BR>MFP (250 ppm F) treated bovine enamel showed a lower reduction rate of Vicker's hardness than non-treated enamel.<BR>In experimental caries in rats, 2000 and 6000 units/ml of dextranase which were administered in drinking water showed a statistically significant reduction of dental plaque, but 5 ppm F of MFP in drinking water did not.<BR>In experimental caries in rats, 2000 and 6000 units/ml of dextranase and 5 ppm F of MFP resulted in the statistically significant reduction of dental caries. Moreover, their combination enhanced the reduction of dental caries more than each agent only. Their cariostatic properties were additive.<BR>These results suggest that the combination of dextranase and MFP which have different cariostatic mechanisms may have an additive effect on the reduction of clinical dental caries.
