Description

It has been shown that Marchantia polymorpha suspension-cultured cells with preculture treatment in a medium containing 0.5 M sucrose acquired a high level of desiccation tolerance. We investigated the tolerance for desiccation of protoplasts isolated from the precultured cells. Survival rate of the protoplasts (0.5 M Sucrose) after desiccation for 48 hrs was above 80%, while that of protoplasts isolated from the cultured cells without preculture (Control) was less than 30%. Furthermore, it was confirmed that the protoplasts have the ability to grow even after desiccation treatment and show almost 80% of cell division rate. We found that the protoplasts isolated from the precultured M. polymorpha cells maintained a high level of desiccation tolerance.
