Breed Variations of Gene Frequencies in Pig Red Cell PHI, 6PGD, PGM and ADA Enzyme Systems and Their Availability for Paternity Test

Abstract

Hemolysates of 464 individuals taken randomly from four pig breed populations (Landrace, Hampshire, Large White and Duroc) in Japan were analysed electrophoreticaly and breed differences of phenotype and gene frequencies in red cell PHI, 6PGD, PGM and ADA enzyme systems were investigated. By using the gene frequencies in each enzyme system, the probability of making exclusions of one of the two possible boars in paternity test was estimated in each breed. PHI, 6PGD, PGM and ADA enzymes were polymorphic in all the breeds investigated and the known alleles were observed in each enzyme system. PHIA and PHIB alleles, in PHI system, were found and all the breeds exhibited a high PHIB frequency. In 6PGD system, a marked difference in 6PGDA and 6PGDB allele frequencies was present among the four breeds. 6PGDA allele had higher frequency in Hampshire and Large White breeds, but lower in Duroc breed. PGMB allele in PGM system showed higher frequency in all the breeds than PGMA allele, and the occurence of PGMA was rare especially in Large White breed. In ADA system, a remarkable difference was observed among the frequencies of four phenotypes in the breed populations investigated. In Landrace and Large White breeds, the three alleles, ADAA, ADAB and ADAO, were present, but ADAB was not present in Hampshire breed and ADAO not in Duroc breed. ADAA was the allele having the highest frequency in Duroc, Large White and Landrace breeds, while in Hampshire breed, ADAO was most frequent. The combined exclusion probability by these four enzyme sytems was estimated to be 0.482 in Landrace, 0.329 in Duroc, 0.326 in Large White and 0.214 in Hampshire breed. Moreover, the combined probability calculated by the eighteen loci (7 blood groups, 5 serum proteins, 2 serum allotypes and 4 red cell enzymes) was 0.926 in Landrace, 0.883 in Large White, 0.829 in Hampshire and 0.814 in Duroc breed.
