The present experiments were designed to clarify the effect of the temperature and duration of supercooling and the cooling rate around 0°C on motility and respiration of bull spermatozoa after supercooled. The results obtained were as follows. 1. Semen was diluted with egg yolksodium citrate solution to contain 0-10% glycerol. When diluted semen was cooled at 1°C/min and supercooled at the temperature range between -2°C and -12°C for 10-60min, motility of bull spermatozoa after supercooled did not decrease in comparison with that of spermatozoa at 4°C irrespective of the temperature and duration of supercooling. To the contrary, when the supercooling state of semen was ceased and semen was frozen at this temperature range, motility decreased with increasing of the duration of freezing. 2. When diluted semen was cooled to-5°C or -10°C at 0.3-80°C/min, sperm motility did not decrease irrespective of the cooling rate in comparison with that of spermatozoa at 4°C as far as they were supercooled. Sperm motility decreased with increasing of the duration of freezing after the supercooling of semen was broken. 3. As far as semen diluted with Seminan, which was a dilutor for frozen semen, was supercooled at -5--10°C for 30min, the oxygen consumption of spermatozoa did not decrease in comparison with that of spermatozoa stored at 4°C regardless of the supercooled temperature and the cooling rate around 0°C. But the oxygen consumption of spermatozoa after frozen at these temperatures decreased with increasing of the duration of freezing. From the results mentioned above, it was suggested that supercooling of semen is not fatal for bull spermatozoa under the present experimental conditions and the death of spermatozoa has a close relationship to the formation of ice crystal of semen.
