Using rat testes of the Wistar strain, the activities of hydroxysteroid dehydrogenases (HSDs) were histochemically examined by the LEVY et al.method, and that of glucose-6-phosphate dehydrogenase (G-6-PDH) by the RUDOLPH and KLEIN method. In the testes 50 days after birth, HSD activities were found in the cytoplasm of only SERTOLI cells and LEYDIG cells as diformazan granules.The activities demonstrated were those ofΔ5-3β-HSD (DHA, pregnenolone and 17α-hydroxypregnenolone as the substrates), 20β-HSD and 17β-HSD (testosterone and estradiol-17β), but none of 20α-HSD.They appeared 20 days after birth in the SERTOLI cells, and reached the maximum on day 30. In the LEYDIG cells, in the meantime, the activities ofΔ5-3β-HSD (DHA and pregnenolone) and 17β-HSD had already existed at birth, while those ofΔ5-3βA-HSD (17α-hydroxypregnenolone) and 20β-HSD came to appear on day 20.The activity of G-6-PDH was always demonstrated where there were any of the HSDs found both in the SERTOLI cells and the LEYDIG cells, the period of their appearance coinciding with that of HSD activities.
