To effect reduction in the cost of production for porcine growth hormone (PGH), use of recombinant BmNPV (Bombyx mori nuclear polyhedrosis virus) in B. mori larvae was envisaged. Accordingly, we constructed an infectious recombinant BmNPV with PGH cDNA inserted into downstream of the polyhedrin gene promoter. The molecular weight of PGH produced by the recombinant virus infected in silkworm cells was 22K daltons, indicating that the cells cleaved the PGH signal sequence at the correct position. The comparison of productivity of the recombinant PGH in mammalian or insect cell lines and in silkworm larvae in in vitro and in vivo showed that the productivity of recombinant protein is about 10×higher in silkworm larvae than that in cell lines. Furthermore, the weight gain assays using hypophysectomized rats confirmed the biological activity of PGH produced in silkworm larvae.
