Abstract

Bovine blood typing monoclonal antibodies have been produced for the purpose of producing unlimited amount of unchangeable reagents in our laboratory.<br>The production of monoclonal antibodies was performed using the standard procedure. BALB/c mice were injected with bovine red blood cells. Myeloma cell lines SP 2, 653 and NS-1 were used. The specificities of monoclonal antibodies were determined with more than 2, 000 red blood cell samples from Holstein breed, Japanese Black, Japanese Brown and Korean native cattle, and also were compared in the 1989 International Comparison Test.<br>The 33 kinds of monoclonal antibodies reacting with antigens in the A, B, C, F, S and Z bovine blood group systems were established. These monoclonal antibodies obtained through the mouse hybridoma technique covered all the polyclonal antibodies that were reported to have been obtained by hetero immunization. And the antibodies reacting with the antigens of the A and F systems were obtained frequently. It can be considered that these antigens are easily recognized as bovine red blood cell antigens in rabbit and mouse.<br>Among these monoclonal antibodies, anti-A2, Fc and Z1 showed quantitative reactions. Antibodies 1 and 24 were named anti-Fc, and they reacted parallel to anti-F1, but hemolysed phenotype V cells at high concentration. By using high concentration Fc antibodies from ascites fluid, it was found that the Fc antigens existed in all of the cattle. Moreover, monoclonal antibody T-3 detected a common epitope of the two S system factors, S1 and U1.<br>These hybridomas continue to secrete antibodies steadily, and the monoclonal antibodies will be useful for bovine blood typing and for the genetic analysis of subtype antigens within a system.
