Effect on plasma cholesterol levels of dietary protein source and methionine deficiency was investigated in chicks fed the 18% protein diet for 10 days. As dietary protein sources, three kinds of alfalfa leaf protein concentrates (LPCs) differing saponin content were used as well as soybean protein isolate (SPI) and casein. Without dietary cholesterol, plasma cholesterol level was always lower in chicks fed the alfalfa LPC diets than those fed the SPI diet. Feeding the high saponin alfalfa LPC significantly decreased both body weight gain and plasma cholesterol level as compared with the LPC with low saponin content. Methionine supplementation at the levels of 0.1% and 0.3% was effective in improving the growth rate of chicks fed the high saponin LPC diet, which was accompanied by the further reduction of plasma cholesterol levels. Body weight gain was also increased by 0.1% methionine addition in chicks fed the low saponin alfalfa LPC and SPI diets, but plasma cholesterol level was unchanged. Addition of 1% cholesterol caused a two-fold increase in plasma cholesterol in chicks fed the casein diet fortified with arginine, methionine and glycine, but it had little effect in those fed the SPI diet fortified with methionine. Plasma cholesterol levels in chicks fed the alfalfa LPC diet were also increased slightly by added cholesterol but the levels were comparable to those fed SPI diet. Substitution of casein with SPI or LPC at the level of 25% and 50% on the crude protein basis suppressed the hypercholesterolemia induced by 1% cholesterol addition, but there was no difference in hypocholesterolemic effect between both plant proteins.
