The present study was undertaken to examine the effect of microtubule (colcemid) and microfilament (cytochalasin B, CB) disrupting drugs on the pronuclear transplantation in the mouse. In the first experiment, enucleation and karyoplast fusion with inactivated Sendai virus were performed between donor and recipient eggs which were pretreated with colcemid (0.1μg/ml), CB (5μg/ml) or colcemid and CB for 15min. In the second experiment, intact eggs at the pronuclear stage were pretreated with colcemid and CB at different conditions to exmine the developmental ability to blastocysts in vitro. In the third experiment, the effect of pretreatment with colcemid and CB for a prolonged period (3hrs) on the pronuclear transplantation was examined. 1) To get high success rates of pronuclear transplantation (95%) and development of reconstituted eggs to blastocysts (66%), the pretreatment of eggs with both colcemid and CB was required (Experiment 1). 2) The treatment of intact eggs with colcemid and CB for 0.5 to 3hrs at 25 to 37°C did not inhibit the development to blastocysts, except for the lowered rate of development in that the eggs incubated with M 16 containing colcemid and CB for 3hrs at 37°C (Experiment 2). 3) Significantly lower proportion of reconstitued eggs developed to blastocysts after pretreatment with colcemid and CB for 3hrs at room temperature as compared with that obtained in eggs pretreated for 15min (53% vs 79%).
